Peregrine Pharmaceuticals Awarded DTRA Contract Worth up to $44.4 Million to Develop Bavituximab for Viral Hemorrhagic Fevers
7/1/2008 7:50 AM - PR Newswire

- Five-Year Defense Threat Reduction Agency Contract Will Support Development of Bavituximab and Fully Human Equivalent as Potential Broad-Spectrum Anti-Viral Agents -

TUSTIN, Calif., July 1, 2008 /PRNewswire-FirstCall via COMTEX News Network/ --
Peregrine Pharmaceuticals, Inc. (Nasdaq: PPHM), a clinical stage biopharmaceutical company developing monoclonal antibodies for the treatment of cancer and hepatitis C virus (HCV) infection, today announced it has entered into a five-year contract worth up to $44.4 million to test and develop bavituximab and an equivalent fully human antibody as potential broad-spectrum treatments for viral hemorrhagic fever infections. The initial contract was awarded through the Transformational Medical Technologies Initiative (TMTI) of the U.S. Department of Defense's Defense Threat Reduction Agency (DTRA).

Bavituximab is Peregrine's lead anti-phosphatidylserine (anti-PS) monoclonal antibody and is currently in clinical trials for the treatment of HCV infections and cancer. In preclinical animal models, bavituximab has demonstrated encouraging anti-viral activity as a potential treatment for viral hemorrhagic fevers. Peregrine's fully human anti-PS antibody, which will also be assessed under this contract, is currently in preclinical development.

Under the terms of the contact, DTRA funds are available to cover testing and development efforts totaling up to $22.3 million over a 24-month base period, with $5 million appropriated immediately for the current federal fiscal year ending September 30, 2008. The remainder of the $22.3 million in funding is expected to be appropriated over the remainder of the two-year base period ending June 29, 2010. The contract can be extended beyond the base period to cover up to $44.4 million in funding over the five-year contract period. Work and funding under this contract are expected to begin immediately.

"This substantial five-year contract award is especially timely in view of recent scientific publications highlighting the broad anti-viral potential of our anti-PS agents such as bavituximab," said Steven W. King, president and CEO of Peregrine. "We are pleased that DTRA has recognized bavituximab's safety profile to date and its promising anti-viral activity, which have been demonstrated in proof of concept models of hemorrhagic fever infection and in two successful clinical trials in patients with chronic HCV infection."

Mr. King continued, "We welcome the opportunity to contribute to the anti-bioterrorism mission of the Defense Threat Reduction Agency, and we also believe that some of the work performed under this contract can be leveraged to support common development tasks applicable to our ongoing bavituximab clinical programs for the treatment of HCV and cancer. The non-dilutive capital provided by this government contract will enable us to further assess the potential of bavituximab to combat the threat of viral hemorrhagic fevers and to help advance the overall bavituximab clinical program."

The DTRA biodefense contract award has the potential to create long-term value for Peregrine, including generating future potential revenues from government stockpiling to combat bioterrorism threats. In the near-term, funding from this initiative will also allow Peregrine to use data and experience obtained from the development and scale-up of bavituximab to support its ongoing clinical development programs.

"We look forward to a close partnership with the DTRA during the performance of this program," said Ronald T. Aimes, Ph.D., associate director of research & development at Peregrine and principal investigator on the contract. "This is an exciting opportunity for Peregrine to contribute to the country's defenses while also achieving important synergies with our development programs for bavituximab and our earlier stage anti-PS technologies."

About Bavituximab

Peregrine's monoclonal antibody bavituximab is the first in a new class of targeted immunotherapeutics that binds to phosphatidylserine (PS), a specific component of certain cellular membranes. PS is normally present only on the inside of cell membranes, but becomes exposed on the external surface of enveloped viruses and the cells they infect. Enveloped viruses are responsible for about half of all human viral diseases, including HCV, influenza, HIV, cytomegalovirus and hemorrhagic fevers. Scientists believe that bavituximab helps block the ability of viruses to infect cells and also helps stimulate the body's natural immune defenses to destroy the virus particles and infected cells. In preclinical studies, bavituximab has demonstrated the ability to bind to a wide range of enveloped viruses and virally infected cells, and it has shown promising activity in animal models of serious viral diseases. In two Phase I monotherapy trials in patients with chronic HCV infection, bavituximab demonstrated encouraging signs of anti-viral activity and appeared safe and well tolerated, with no dose-limiting adverse events. A clinical trial of bavituximab for the treatment of HCV patients co-infected with HIV is ongoing. Bavituximab is also in multiple Phase II trials for the treatment of solid cancers.

About the Defense Threat Reduction Agency

The Defense Threat Reduction Agency (DTRA) was founded in 1998 to integrate and focus the capabilities of the Department of Defense that address the weapons of mass destruction (WMD) threat. The mission of the DTRA is to safeguard America and its allies from WMD (e.g. chemical, biological, radiological, nuclear, and high yield explosives) by providing capabilities to reduce, eliminate, and counter the threat, and mitigate its effects. Under DTRA, Department of Defense resources, expertise and capabilities are combined to ensure the United States remains ready and able to address the present and future WMD threats.

Peregrine Pharmaceuticals Highlights Promising Early Data From Its Three Phase II Bavituximab Cancer Trials
8:02a ET June 3, 2009 (PR NewsWire)
Peregrine Pharmaceuticals, Inc. (Nasdaq: PPHM) today highlighted the progress that the company has achieved in its Phase II program assessing the combination of bavituximab and chemotherapy in three separate cancer trials. Bavituximab is a monoclonal antibody with a unique mechanism that allows the body's own immune system to recognize and act on the tumor and its supporting blood vessels, resulting in anti-cancer effects. Bavituximab is currently being tested in combination with chemotherapy in one Phase II trial in advanced lung cancer and two Phase II trials in advanced breast cancer. Recently-reported data highlights from these studies include the following:

-- Non-Small Cell Lung Cancer: Bavituximab in Combination with Carboplatin and Paclitaxel In the Phase II trial evaluating bavituximab in combination with carboplatin and paclitaxel in non-small cell lung cancer (NSCLC) patients with locally advanced or metastatic disease, 11 of 17, or almost 65% of evaluable patients in the initial cohort of 21 patients achieved an objective tumor response. These early results, which exceeded the pre-specified endpoint needed to expand the trial, compare very favorably with historical data with chemotherapy alone and are especially encouraging in this hard-to-treat cancer. Patient enrollment and dosing are continuing in the expansion stage of the trial, which will enroll an additional 28 patients for a total of 49 NSCLC patients overall. -- Advanced Breast Cancer: Bavituximab in Combination with Docetaxel In the Phase II trial evaluating bavituximab in combination with docetaxel in advanced breast cancer patients, enrollment of the planned 46 patients was recently completed. As reported in an oral presentation at the 2009 ASCO Annual Meeting, 10 of 14, or 71% of evaluable patients in the initial 15-patient cohort demonstrated an objective tumor response. These data exceeded the pre-specified endpoint needed to expand the trial and compare very favorably with historical data with chemotherapy alone. Recent analysis also shows the median progression free survival of the patients enrolled in the first part of the study was 7.4 months, an additional promising early result. Patient dosing and follow-up in this trial are continuing. -- Advanced Breast Cancer: Bavituximab in Combination with Carboplatin and Paclitaxel In the Phase II trial evaluating bavituximab in combination with carboplatin and paclitaxel in advanced breast cancer patients, nine of 14, or 64% of evaluable patients in the initial 15-patient cohort achieved an objective tumor response. These data exceeded the pre- specified endpoint needed to expand the trial and compare favorably with historical results with chemotherapy alone. Patient enrollment and dosing are now underway in the expansion stage of the trial, which will enroll an additional 31 patients for a total of 46 advanced breast cancer patients overall.

The primary objective of these multi-center, open-label studies is to assess the overall patient response rate to the combination regimen of bavituximab and chemotherapy according to RECIST criteria. Secondary objectives include measuring time to tumor progression, duration of response, overall patient survival and safety parameters. The Phase II bavituximab cancer trials have a Simon two-stage design, where an initial cohort of patients is treated and evaluated and then the study is expanded to a second larger cohort of patients if pre-specified criteria are met. All three trials surpassed the criteria for expanding enrollment to the second cohort. Enrollment of the expanded cohort is now complete in one trial and is proceeding in the other two trials.

"Having just completed a busy round of successful data presentations and partnering meetings at ASCO, we are gratified at the level of interest the bavituximab program is now receiving from leading cancer researchers and drug developers," said Steven W. King, president and CEO of Peregrine. "We believe this growing interest reflects our recent progress in successfully advancing all three Phase II bavituximab cancer trials, easily surpassing the pre-determined efficacy criteria needed to expand the trials to larger patient cohorts. The trials encompass different cancers and different chemotherapy regimens, yet in all three, preliminary data on tumor responses are very encouraging and compare well with historical experience with chemotherapy alone. With these encouraging results in hand, we look forward to sharing additional data on the entire clinical study populations in the second half of 2009."

Bump. On watch for a bounce, dropped from 95 cents earlier this month to a 69 cent close on Friday. IMO 60 cents would be a great entry, 65 cents if you have a high risk tolerance. Then if the 200DMA at 56 cents doesn't hold abandon ship....

UPDATE 1-Peregrine Pharma lung cancer trial shows promise
10/5/09 8:42AM

* Lung cancer drug improves progression-free-survival rate

* Shares up 14 pct in premarket trade

Oct 5 (Reuters) - Peregrine Pharmaceuticals Inc (PPHM.O) said a combination of its experimental cancer treatment with chemotherapy improved progression-free-survival in lung cancer patients as compared to chemotherapy alone in a mid-stage trial.

The trial is testing the overall response rate to Peregrine's bavituximab -- a monoclonal antibody -- in combination with chemotherapy in patients with non-small cell lung cancer.

Monoclonal antibodies are tailor-made proteins that can detect the proteins attached to cancer cells and be used to block their growth.

An analysis from a 21-patient cohort of the trial showed the median progression-free-survival for patients receiving bavituximab and chemotherapy was 6.5 months, compared to a survival rate of 4.2 months to 4.5 months for those receiving only chemotherapy.

Peregrine also said that it completed enrolling the total of 49 patients planned for the study.

Lung cancer kills about 1.2 million people a year and is the top cause of cancer death globally.

Shares of the company were up 14 percent to $0.80 in premarket trade. They closed at $0.70 Friday on Nasdaq

If a mod sees this, please change thread to PPHMD...

Ticker changed from PPHM to PPHMD after a 1:5 reverse split took affect today. Gap filled and came below the 200dma just where I wanted to get in...I saw it down a good 5% today and decided to wait. I checked the pps after hours and it ended up closing up 6%!

Here is the chart:
